摘要:
    Vigilin是一个由14个连续的KH结构域组成的RNA结合蛋白。它可以通过与mRNA结合调控mRNA的翻译后表达水平,并能够结合脂蛋白。本实验室此前的实验中发现Vigilin参与了对雌激素信号传导通路的调控,为详细研究其调控机制,文章通过凝胶阻滞实验证明了Vigilin能在体外结合于雌激素受体3非翻译区(ERbeta 3UTR)。借助双荧光素酶报告基因实验发现,高表达Vigilin可以提高雌激素信号通路中的重要受体ERbeta的翻译效率。利用激光共聚焦显微镜和荧光蛋白定位实验,发现Vigilin在细胞内大量定位于pbody中。通过上述实验证明了Vigilin能够通过结合ERbeta 3UTR调控ERbeta的表达量,参与了细胞内雌激素信号传导通路。由于ERbeta是雌激素信号传导中非常重要的受体,因此该研究具有重要的意义。
