摘要:
    用EcoR I和Sal I将目的基因片段6His11ReGFP从载体pDC3166His11ReGFP上酶切下来,并将该目的基因片段与供体载体pFastBac1连接构建供体质粒pFastBac16His11ReGFP;将其转化大肠杆菌(Escherichia coli)DH10Bac感受态细胞,经卡那霉素、四环素、庆大霉素及蓝白斑筛选得到重组杆状病毒载体pBac6His11ReGFP,PCR鉴定得到单一目的基因条带。将该病毒载体转染Sf9细胞,进行病毒包装,在倒置荧光显微镜下观察蛋白表达,并以荧光定量PCR方法测定病毒滴度。
