摘要:
为了探究黄瓜花叶病毒(Cucumber mosaic virus, CMV)复制酶2a蛋白细胞核定位的生物学意义,以本氏烟UNE12蛋白(NbUNE12)为研究对象,通过双分子荧光互补实验及病毒诱导的基因沉默方法分析该蛋白与2a蛋白的相互作用情况及其在CMV侵染过程中的作用。结果表明:CMV 2a蛋白与NbUNE12蛋白的相互作用主要集中于细胞核中;下调表达NbUNE12增加CMV的2b缺陷型突变体(CMVΔ2b GFP)在植物系统叶的病毒含量,表明NbUNE12蛋白能够与CMV 2a蛋白发生相互作用,并且负调控CMVΔ2b GFP侵染本氏烟。该研究为CMV 2a蛋白在植物细胞核定位的生物学功能进一步研究奠定理论基础。
