摘要:
为了探究转录因子Runx3在人外周血T细胞中的功能,通过检测T细胞亚群中该基因的表达差异,并在过表达Runx3基因后分析T细胞亚群分布,进一步比较抑制性受体和下游基因的表达差异。结果表明:Runx3主要表达于初始T细胞亚群中,与对照组相比,过表达Runx3可显著提高T记忆干细胞比例,且PD1、Tim3以及Lag3的表达显著降低;RTqPCR结果表明,Runx3T细胞中Blimp1表达显著降低,SIRT1 表达显著升高。因此,Runx3蛋白可能通过调控T细胞分化和维持记忆表型相关基因 Blimp1和SIRT1来提高T记忆干细胞比例,从而能够维持T细胞功能,延缓其衰竭。研究结论可为改进T细胞免疫疗法提供了重要的理论依据。
