摘要:
大多具有分支的β-(1-3)-D-葡聚糖具有免疫调节及抗肿瘤活性。文章利用人单核白血病THP-1细胞株建立细胞水平上葡聚糖生物活性的检测方法,实现高通量筛选活性多糖。THP-1细胞在不同浓度的葡聚糖刺激48h后离心收集上清,ELISA法检测其释放细胞因子浓度,以筛选有效的评价指标。结果显示:THP-1细胞在不同葡聚糖刺激下,细胞培养液中IFN-α含量变化明显。IFN-α可作为在细胞水平上评价葡聚糖活性的标准检测指标。该方法既克服了动物实验工作量大、成本高,又克服了原代细胞不稳定等因素,为高通量筛选活性多糖提供了新途径。
