摘要:
     DNA甲基化异常在肿瘤发生中起着极为重要的作用,而基因组DNA的甲基化主要通过DNA甲基化转移酶(DNA methyhransferases,DNMTs)来完成。microRNA和甲基化转移酶的表达具有一定的联系。构建携带有miR29b的腺病毒载体并包装病毒,鉴定正确后,将病毒感染肝癌细胞PLC,用qRTPCR的方法检测miR29b在PLC细胞中对甲基化转移酶表达的调控作用。结果表明,成功构建携带有miR29b的腺病毒,qRTPCR方法检测表明miR29b能有效地调控甲基化转移酶的表达。
