摘要:
猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)是造成猪腹泻的一种主要肠道病毒。为建立一种能快速、准确和适于大批量样品的检测PEDV方法,根据PEDV N蛋白基因保守序列特征设计了一对特异性引物,并对退火温度和循环数等条件进行了优化。结果显示:建立的RTPCR方法特异性地扩增出PEDV目的条带,对非靶标病毒均无交叉反应,最低检测限可达5拷贝/μL;应用该方法对浙江40份健康猪粪便样品进行检测,PEDV检出率为12.5%。结果表明,该文所建立检测PEDV的方法具有较高的特异性和灵敏度,可用于临床诊断和流行性病学调查。
