摘要:
药物洗脱支架(Drugeluting stent, DES)释放抗增殖的药物来抑制平滑肌细胞(Smooth muscle cells, SMCs)增殖,阻止内膜增生。但这些药物不可避免地会延迟内皮化,因此有必要寻找合适的药物选择性的抑制SMC增殖,同时促进内皮细胞(Endothelial cells, ECs)增殖。维生素C(Lascorbic acid, LAA)是一种用于口服或是直接加入细胞培养的药物,因此实验中在培养过夜的EC和SMC细胞中加入浓度为300 μg/mL的LAA、浓度为100 μg/mL 的雷帕霉素(Sirolimus,SIR)、高糖培养基(Dulbeccos modified eagle medium, DMEM)、杜氏磷酸缓冲液(Dulbeccos phosphate buffered  saline, DPBS)和乙醇各100 μL,培养7 d观察,结果表明:SIR能够抑制EC增殖,而LAA能够显著促进EC的增殖,同时也能够抑制SMC的生长;在培养过夜的EC和SMC细胞中加入100 μL不同浓度的LAA(1、100、300、500 μg/mL和1000 μg/mL),培养7 d后,1、100 μg/mL和300 μg/mL的LAA有利于EC增殖,而500 μg/mL和1000 μg/mL的LAA会抑制EC增殖,并且300 μg/mL和500 μg/mL的LAA可以显著抑制SMC增殖。由此表明维生素C可以成为潜在的用于药物洗脱支架的药物。
