摘要:
为评估纳米羟基磷灰石颗粒负载p53 基因对肝癌干细胞的抑制效果,采用化学沉淀法制备羟基磷灰石,对其进行表征,并检测其生物相容性。通过成球培养法获得肝癌干细胞,用经聚乙烯亚胺(PEI)修饰的纳米羟基磷灰石颗粒(HP)负载p53基因(HP p 53)处理肝癌干细胞,用MTT法、hoechst 33258染色综合评估杀伤效果。结果表明:所制备的纳米颗粒主要成分和晶型为羟基磷灰石,呈短棒状,长径为20~50 nm,短径约为20 nm,并具有良好的生物相容性和降解性;HP p53处理后的肝癌干细胞存活率显著下降;用Hoechst 33258染色观察到HP p53处理后的肝癌干细胞细胞核出现高亮光斑,细胞密度降低,表明所制备的纳米羟基磷灰石负载p53基因对肝癌干细胞具有显著的抑制效果。
