摘要:
雌激素在体内的生理作用通过雌激素受体(Estrogen Receptor)ERβ和ERα的介导来实现调控。ERβ在乳腺癌、前列腺癌、结肠癌和卵巢癌的发生、发展过程中具有调控作用。ILF3是RNA结合蛋白中的一员,参与细胞周期调控。凝胶阻滞实验(EMSA)证明了ILF3蛋白能结合ERβ mRNA 3 UTR。雌激素受体可介导雌激素实现其功能,ILF3蛋白可能通过与ERβ结合参与雌激素信号通路的调控。为探讨他们的关系以及调控机制,构建ILF3过表达的慢病毒载体,通过Western blot检测ERβ蛋白的表达情况。在蛋白水平上,ERβ蛋白的表达量随着ILF3蛋白量的升高而升高,ILF3对ERβ起正向调控。MTT实验检测ILF3基因表达对MCF 7细胞活力有影响。
