摘要:
端粒是真核生物线性染色体末端的特殊结构,一定长度的端粒在维持真核生物遗传信息完整性和染色体结构稳定性中起重要作用。建立一种简单快捷定量测定水稻单个端粒绝对长度的方法对研究水稻各个染色体端粒具有积极的意义。研究将特殊接头连接到水稻染色体端粒的Goverhang末端,并利用近端粒区域的特定引物以及下游接头引物,通过PCR技术,获得所需鉴定的端粒,结合DNA凝胶电泳成像分析系统,定量检测水稻单个端粒的长度。结果显示本方法可以通过少量水稻基因组DNA有效测定单个端的粒长度。
