摘要:
    将掌叶半夏凝集素和死亡素的融合基因〖WTBX〗thanatin\|linker\|ppa〖WTBZ〗定向连接到pPIC3.5K,构建酵母表达载体pPIC3.5K/Thanatin\|(G8S2)\|PPA并进行序列分析,经〖WTBX〗Sal〖WTBZ〗I单酶切线性化后电转化到GS115毕赤酵母感受态细胞中,PCR鉴定后在含有G418抗性的YPD平板筛选高拷贝重组子。利用甲醇在BMMY培养基中诱导表达融合蛋白,经SDS\|PAGE和Western bloting分析显示获得分子量约31kD的融合蛋白,融合蛋白经纯化后MTT分析其抗肿瘤活性,结果显示融合蛋白具有显著的抗肿瘤效果。
