摘要:
    利用改造后的溶瘤腺病毒Ad.enAFPE1A△E1B55Kd作为载体携带治疗基因MnSOD,得到目的病毒Ad.enAFPE1A△E1B55KdMnSOD;通过PCR方法鉴定病毒构建正确;MTT法检测病毒对肝癌细胞的特异杀伤能力和对肝正常细胞的安全性;Hoechst33342染色实验观察细胞凋亡的现象。结果显示:经目的病毒感染后肝癌细胞出现明显的病变现象和生长抑制,而肝正常细胞基本无病变现象出现,表明构建的目的病毒具有较高的安全性和很好的靶向性。此外目的病毒处理能引起特异性的肝癌细胞凋亡,所携带治疗基因MnSOD能通过促进肿瘤细胞的凋亡达到抑制肿瘤细胞生长的效果
